A three-times weekly service will operate to Mumbai, starting on 19 December, with a twice a week service to Delhi taking off on 5 January.
Bookings for both 787 services open on October 20.
The carrier already operates daily Heathrow services to both cities.
With India still in the grip of Covid, the flights are expected to largely cater for the visiting friends and family market.
Virgin Atlantic Chief Commercial officer Juha Jarvinen said: “Following the relaunch of services from Heathrow to Mumbai and Delhi in September 2020, these new services from Manchester represent our continued investment in India.
“Both Mumbai and Delhi are popular year-round destinations, and we look forward to welcoming travellers from the North and Midlands onboard as demand for leisure and business travel gradually increases to the region.”
Virgin Atlantic is resuming its Manchester to Barbados service on 26 October, followed by its first flight from Manchester to Islamabad on 10 December.
